摘要
本文研究用IFN_ 和CSF联合LPS诱导和调节小鼠骨髓及腹腔巨噬细胞产生CF。其中用IFN_ 联合LPS诱导的BMC培养上清对L9219细胞的溶解杀伤作用高于其它组.rHIFN_ 与LPS对诱生小鼠BMC产生CF有协同作用;细胞毒因子产生量均以培养4h为最佳;BMC为体外诱生CF的较敏感细胞,适于分析各种诱导物体外诱导CF的产生效果.
Interferon(IFN) and colony-stimulating factor (CFS) with lipopolysaccharide (LPS) were used to induce and regulate murine bone marrow cell (BMC) and peritoneal exudate cell (PEC) to produce cytotoxin factor (CF) in this study. The lysis and killing effect of supernatants from this cell cultures on L929 cell were analysed. The cytotoxicity of the group of LPS+rHIFN_α was significantly higher than that of other groups. The results showed that:rHIFN_α and LPS had syergism effect in inducing mouse BMC to produce CF; the highest level of CF production in all of the 3 treated groups were obtained at the time of 4 h culture; BMC appears to be a kind of more sensitive cell which can produce CF in vitro. This assay system is suitable for the research of evaluating the ability of the various inducers to produce CF in vitro.
