摘要
用EDC法将ABEI标记于Fg上,不影响Fg转变成F及F对t-PA的结合能力。ABEI-Fg固相后测定对F有结合能力和对Pg有激活活性的t-PA。用这个方法。t-PA在0.078…40IU/nì范围,浓度对数值与发光计数值呈直线正相关,灵敏度比溶解圈法高30倍以上。检测样品批内水平均变异系数6.63%,批间平均变异10.27%,样品中加入t-PA标准的平均回收率98.57%,样品液中加入40mM6—氨基已酸能抑制非特异蛋白水解酶对固相F-BBEI的非特异水解,不影响t-PA与F结合。
When the Fg was marked by ABEI with EDC method the combining capability of F for t-PA was not influenced After the solid phase of ABEI-Fg was done the t-PA that had comdining capability for F and actinated activity for Pg was determined In the range of 0.07-40IU/ml a good linear dorrelation was found between the logarithmic numerical value of t-PA concentration and luminescent numerical value In thess samples the coefficient of variationsof intra-and inter-assav were 6.63% and 10.27%respectivel y. The recowery rates of the samples which the t-PA was put in were 98.5% The non-specilic hydrolysis of the solid phase FABEI which might be hydrolyzed by the non-spedific prbteolytic enzyme was inhibited if the 40mM of 3-Aminocaproic Acid was put in the samples and the combining of the t-PA and the F would not be influenced.
