黑曲霉果胶酶的研究Ⅲ工厂试验及酶作用方式

摘要 通过实验室和工厂试验,证明603菌株培养条件简单,生产周期短、酶活性高,是一株有生产价值的菌株。用CM-Sephabex C-50 Sephadex G-75提纯果胶酶,得六个园盘电泳均一性组份。对四个组份进行了酶学性质的初步研究,它们的最适作用温度和pH为:4-G-1 60℃、pH4.0~4.2;4-G-2 50℃,pH4.4-4.6;2-GC-2 50℃,pH3.6;2-GC-3 40℃ pH4~4.4。Km值分别为:0.446;0.392,0.274,1.634。层析图谱证明:4-G-1和4-G-2是内切酶,2-GC-2和2-GC-3是外切酶。 Through laboratory and factory experiments we confirm that, the strain 603 has simple culture condition, short producing cycle and high enzyme activity. It is one strain of producing value. We purify pectic enzymes with CM-Sephadex G—50 and Sephadex G—75 and get six uniform disc electrophoresid components. Prelimitary study on the four of them in enzymology properties their suitable factors are: 4—G—1 60℃ pH4.0—4.2, 4—G—2 50℃ pH 4.4—4.6, 2—GC—2 50℃ pH3.6, 2—GC—3 40℃ pH 4.0—4.4.Km. is respectively 0.446, 0.392, 0.274, 1.634. The chromatogram proves that 4—G—1 aod 4—G—2 are endo-enzymes, 2—GC—2 and 2—GC—3 are exo-enzymes.
