摘要
本文建立了利用高效液相色谱法定量分析L-山梨糖发酵过程中主产物2-酮基-L-古龙酸含量的方法。色谱柱为Nuclesoil-N(CH_3)_2 5μm;流动相为0.01M KH_2PO_4,用2%H_3PO_4调到pH=4.0,流速为1ml/min,检测器为示差折光仪(RI)。在该条件下色谱峰面积与进样的2-酮基-L-古龙酸含量之间呈线性相关,相关系数R=0.9965,最小检出量为0.1%。检测结果与采用碘量法分析结果一致。经多次使用,证明该方法简单可靠,能准确及时地判断L-山梨糖发酵的终点。
An improved method to assay keto-2-L-gulonate in fermentation processes with HPLC has been established. The chromatography condition is as follows. Column is packing with Nuclesoil-N (CH_3)_2 5μm, mobile phase 0.01M KH_2PO_4 and adjust pH=4 with 2% H_3PO_4 flow rate 1m1/min, detector RI-2H differential refractometer. The peak area shows linear relationship with the contents of keto-2-L-gulonate in samples, R=0.9965. The results with HPLC are in accordance with that of iodimetry and can be used in industrial fermentation analysis.
