摘要
本文报道了A组RV McAb的特性鉴定及临床初步应用。用从河南省腹泻犊牛粪便中分离的HN-7毒株(RV)免疫BALB/C小鼠,用杂交瘤技术获得2株分泌A组RV特异性McAb的杂交瘤细胞。所获2个McAb(2C_7、3C_9)与A组RV第1血清型Wa株、第3血清型SA-11株、第6血清型NCDV株,以及从国内猪、牛粪便中分离的4株未分型毒株的间接ELISA效价达1:10~5~1:10~6,但与这7个毒株的血凝抑制滴度≤1:8,中和效价≤1:100,表明这2个McAb是针对A组RV共同抗原的。用此McAb建立的单夹心ELISA技术,测定162人、牛腹泻粪样,与常规ELISA总符合率为98.8%。
Abstract The characterization and application of the specific McAbs against rotavirus A were reported in this paper. Two McAbs(2C7 and 3C9)were obtained by fusion of SP2/0 myeloma cells with spleen cells from BALB/c mice immunized with rotavirus of HN 7 strain isolated from diarrheal calf in Henan Province. Both the McAbs(2C7, 3C9)reacted with the groupcommon antigen of group-A RV, because the titer of two ascites McAbs had a titer of 10^(-5)~10^(-6) by indirect ELISA with RV of bovine HN-7, BRV007. BRV014, NCDV, P86, SA-11 and Wa strains; but they had titer 2^(-3) and 10^(-2) respectively by HI and NT. According to result of Ouchter-lony gel diffusion test, 2C7 McAb was identified us mouse IgGl subclass. The cor-respondent rates of single-sandwich ELISA and routine ELISA detecting RV anti- gen in fecal samples of diarxheal calfes and human babies were 98.8%(160/162).
