摘要
为了满足对单克隆抗体的需要,我们应用小型发酵罐对杂交瘤细胞进行了培养试验,并对可能影响细胞生长和抗体产生的因素作了初步探讨。同时还对降低血清含量对培养杂交瘤细胞的影响进行了试验。结果表明,3株细胞在发酵罐中生长良好。细胞最大密度达8×10<sup>2</sup>/ml以上。培养上清中的抗体滴度接近静止培养水平,其中A<sub>6</sub>和A<sub>11</sub>】10<sup>5</sup>,LS<sub>7.161</sub>】1:64。接种细胞密度、培养时的pH、溶氧等对细胞生长有不同程度的影响。驯化的细胞在发酵罐中逐步降低血清量是可行的。
We have done hybridoma cell culture with 3.5L fermenter and investigated some factors, such as pH and pO_2 which might influence cell propagation and secretion of antibody. Moreover, we have tried to cultivate the cell with lower concentration of FCS media.
The results showed that three cell lines could grow well in fermenter. The maximum cell density was more than 8×10~5/ml . The antibody titre in supernantant was almost equal to that obtained by flask cultivation. Titres of cell lines A_(?) and A_(?) was greater than 10~5 and LS_(7.161) were greater than 1:64. The cell density of inoculation, pH and pO_2 showed various effects on cell growth. Gradually lowering the FCS concentration during cultivation in fermenter would be possible.
