抗 A、B 和 C 群脑膜炎球菌多糖单克隆抗体的制备及鉴定

Production and Characterization of Monoclonal Antibodies against Meningococcal Polysaccharides Groups A,B and C

10株抗 A、B 和 C 群脑膜炎球菌多糖 McAb,经 PHA、PHAI、ELISA 和试管凝集试验测出较高的群特异性抗体滴度。经中国药品生物制品检定所选用12个群脑膜炎球菌国家标准菌株和地方菌株以及其它奈瑟氏菌株进行玻片凝集和试管凝集检定。结果显示 McAb 滴度高、特异性强,用于分群诊断取得满意的结果。10株 McAb 均属小鼠 IgM 类。

Several monoclonal antibodies(McAbs)with specificity for sero groups A,B and C Polysaccharides have been produced. The serogroup specificity,titer,affinity,stability and mouse Ig subclass of these McAbs were determined with passive hemagglutination test,hemagglutination inhibiti- on test,enzymelinked immunosorbent assay,slide agglutination test,quantitative agglu- tination assay and double diffusion. The results showed high titer and serogroup specificity.When tested on 39 and 41di- fferent serogrouping N.meningitidis strains and other Neisseria species,these McAbs were able to specifically agglutinate standard strains as well as isolated strains from different regions.These McAbs were identified as mouse IgM.
