摘要
本文用毒理学研究手段,探讨用过期四环素致肾脏损害的动物模型的制作方法及观察手段,为进一步研究肾脏损害,特别是肾小管酸中毒的发病机理,病理改变、临床表现、诊断及治疗打下基础。本项研究用过期四环素针(粉)剂,以每日0.5mg/10g、每日1mg/10g、每日2mg/10g的剂量给小鼠灌胃,按不同累积摄入量分5个组。观察指标有:血气分析,尿气分析,病理及超微结构的变化等。通过形态学观察证实,过期四环素能制出肾脏近曲小管损害的动物模型,初步观察到过期四环素对肾脏超微结构效应时相,并提出小鼠的血气分析、尿气分析资料。
In this artical, kidney injury by overdue tetracycline was studied with the method of toxicology so as to lay foundations of further study renal tubular acidosis. According to the different average accumulative intake of overdue tetracycline, experimental mice were divided into five experimental groups and one control group freely, blood pH,blood gas, urine PH and urine gas were examined.The blood pH was form 7.217 to 7.271 and no statistical significant defference was found between these groups. There was highly Positive correlation between the values of pH, BB and HCO_3 in urine and the different average accumulative intake of overdue tetracycline. All the epithelial cells of the renal Proximal tubules became cloudy swelling and the cavity narrow in experimental group 4 and 5.Electron microscopy disclosed that the mitoehondria were swelling or its cristae disappearance in group 1 and 2. We also found that the effective Phase of overdue tetracycline to dameage mitochonrial ultrastructure was different from mercury bichloride. On the basis of the results,the authors believe that the effect of overdue tetracycline on the mitochondiral damage may be as starting factor causing injury to the kidney.
