大鼠D-氨基半乳糖肝损伤后肝细胞几种组织化学定量分析及血清学观察 被引量:5

QUANTITATIVE HISTOCHEMICAL AND SEROLOGIC STUDIES ON RAT LIVER DAMAGE INDUCED BY D-GALACTOSAMINE

摘要 本实验用国产D-氨基半乳糖诱发大鼠肝损伤后对肝细胞的DNA、RNA和肝糖原含量及葡萄糖—6—磷酸酶、琥珀酸脱氢酶的含量用4种不同类型的图象分析仪进行了原位定量分析,并观察和分析了血清学改变。实验结果表明,损伤组大鼠肝细胞的DNA、RNA和肝糖原含量明显低于正常对照组,葡萄糖—6—磷酸酶和琥珀酸脱氢酶活性明显下降,血清谷丙转氨酶、总胆红素、硷性磷酸酶、甘油三酯、游离脂肪酸均升高,血糖下降。本文对D-氨基半乳糖诱发肝损伤的机理及用图象分析仪对组织化学结果进行定量分析等问题进行了讨论。 Rat hepatitis induced by D-galactosamine (D-Gal) was studied with histochemical techniques & serum components, the quantitative analyses on the histochemistry results were made with 4 different analysers. Experimental proceeding:(1) 30 Wistar rats were divided into 2 groups, D--Gal group & normal group; (2) Routine histochemistry was carried out; (3) contents of DNA & RNA in hepatocytes were determined with Quantimet 970 image analyser; (4) Activity of glucose-6-phosphatase (G-6-Pase) with MPV Ⅲ-TAS PLUS; (5) Activity of succirate-dehydrogenase(SDH) with MAGISCAN Ⅱ; (6) Liver glycogen with MICROVIDEOMAT; (7) Serum components were determined with HITACHI 705 blood autoanalyser that include GPT, tatal serum bilirubin (T-BIL), triglycerides (TG), free fatty acids (FFA), alkaline phosphatase (ALP) & glucose. Results: In the G-GAL group, Contents of DNA, RNA & liver glycogen were significantly reduced (P<0.05); Activities of G-6-Pase & SDH were weaker than that of the normal (P<0.01). The level of GPT rose to 22.9 times the normal value, T-BIL, TG & FFA were 4 to 5 times higher than that of the normal, ALP also showed a pronounced increase (P<0.01), however glucose decreased (P<0.05). The above alterations were similar to human viral hepatitis.
