动物实验表明人体器官内淋巴郁滞可引起许多疾病或使其恶化。本研究采用0.05ml^(125)I标记的人血浆r球蛋白(5g/dL,185KBq,5μCi)兔肝被膜下注射,电子计算机数字闪烁出现两种清除参量,快速清除参量与血流对同位素的吸收有关,慢速清除参量与淋巴循环和蛋白分解有关。在正常对照组11只兔中,慢速清除平均半清除时为288分钟(标准误24.7)。在10只胸导管结扎的兔中,24小时后肝脏发生中度水肿,慢速清除参量显示半清除时延迟到平均525分,与正常对照组相比,有极为显著的统计学意义(P<0.001,标准误19.1)。然而也说明,胸导管虽然结扎,仍有部分淋巴液回流。胸导管结扎所引起的这种中度淋巴郁滞的实验模型,与人体许多疾病所引起的淋巴郁滞相类似。在另外9只兔也行胸导管结扎,并在结扎后立即腹腔注射香豆素(25mg/kg)。这种药物可以从组织中通过蛋白的分解作用移除过多的蛋白质。24小时后同样方法测定,本组动物的半清除时为286分(标准误12.8)。与正常对照组相比,没有差异性。但是与没有用药的胸导管结扎组相比,却有非常显著的统计学意义(P<0.001)。本实验结果提示放射性同位素的示踪研究,可应用于器官内淋巴的循环测定,同时证实了香豆素对肝脏及其它内脏器官有关疾病的治疗作用。
Many animal experiments have shown that lymphostasis of human organ probably causes or aggrevates. many diseases. In the present study 0.05 ml of (125)~I-labellcd human gammaglobulin(5g/dl, 185KBq (5μCi)was injected just beneath the surface the livers of rabbits. The disappearance curve had two components, they are the fast one which clearance with uptake by blood, and the slow component reflected clearance by the lymph and tissue proteolysis. The mean half-clearance time (1/2t) of the slow cornponcnt for 11 normal rabbits was 288 minutes (S. E. 24.7). The thoracic ducts were ligated in 10 rabbits. There was evidence of middle lyrnphostasis in the liver after 24 hours. The mean half-clearance time was very significantly (p<0.001) increased to 525 minutes (S.E19.1). It was, however, shown that the ligating the thoracic duct still permits some lymph flow. This model of moderate lymphostasis was selected to simulate a human disease. In addition, in nine rabbits the thoracic ducts were also ligated, and at that time, they were given a intraperitoneal injection of 25 mg/kg of coumarin(5,6-Benzo-[alph]-pyron). This removes excess protein from the tissues via increased proteolysis. Twenty-four later, the mean half-clearance time in these animals was 286 minutes (S.E. 12.8). This was not significantly different from that of the normal animals, but very different (P<0. 001) from the ligated, untreated animals. The result suggested that the radio-isotope trace study could probably be used to meassure the lymph circulation of the organs. At the mean time, there was effective action of coumarin on the related diseases in the liver or other orans.
