补肾填髓方对去势兔骨髓间充质干细胞钙化结节、骨钙素作用的研究 被引量:2

摘要 目的:探讨补肾填髓方促进去势兔骨髓间充质干细胞(MSC)s的成骨分化能力。方法:应用Van Kossa染色法和ELISA法对比补肾填髓方和成骨诱导剂对MSCs钙化结节和骨钙素的影响。结果:补肾填髓方能显著提高去势兔MSCs钙化结节数量,与去势组比较,差异有非常显著性意义(P<0.01),但作用较成骨诱导剂弱(P<0.05),同时也能显著提高骨钙素含量,与去势组比较,差异有显著性意义(P<0.05),钙化结节数量和骨钙素含量的增多与补肾填髓方用药时间呈正相关(P<0.01)。结论:补肾填髓方用于治疗骨质疏松症,与促进MSCs向成骨细胞分化相关,但其促进MSCs成骨的机制仍需进一步研究。 Objective: To investigate the effect of Bushen Tiansui Capsule on osteogenic differentiation of rabbit bone marrow mesenchymal stem cells (MSCs). Methods. Van Kossa staining method and enzyme linked immunosorbent assay (ELISA) were applied to investigate the osteogenic differentiation effect of Bushen Tiansui Capsule and osteogenic derivatives on the formation of calcified nodules and osteocalcin(OCN) content in ovariectomized rabbits. Results: Bushen Tiansui Capsule obviously elevated the number of calcified nodules and the content of OCN in MSCs of ovariectomized rabbits (P 〈 0.05 or P 〈 0.01), but the effect was weaker than that of'osteogenic derivatives(P 〈 0.05). The number of calcified nodules and the content of OCN were positively correlated with the medication time of Bushen Tiansui Capsule (P 〈 0.01). Conclusion, Bushen Tiansui Capsule can treat osteoporosis in clinically by improving osteogenic differentiation of MSCs. Its mechanism in improving the osteogenesis of MSCs still needs further research.
