葛根提取物对高尿酸血症模型大鼠尿酸及血脂的影响 被引量:15

摘要 [目的]研究葛根提取物对高嘌呤饲料饲喂致高尿酸血症模型大鼠的降血尿酸及调节血脂作用。[方法]SD大鼠90只,随机分为正常对照组、模型对照组、葛根水提液三个剂量组(5.25g/kg、3.50g/kg、1.75g/kg)、葛根醇提液三个剂量组(5.25g/kg、3.50g/kg、1.75g/kg)、阳性对照组(别嘌醇0.01g/kg)。采用连续饲喂高嘌呤饲料25天建立大鼠高尿酸血症模型,每天在给予高嘌呤饲料造模的同时给药,连续25天。试验期间,观察大鼠一般行为学、体重;给药第8天、第15天分别眼眶取血测定血清尿酸(UA)、总胆固醇(TC)、甘油三酯(TG)含量;末次给药1h后眼眶取血测定血清UA、TC、TG含量,血液流变学指标。[结果]①造模第8天,葛根醇提剂量组(5.25g/kg、3.50g/kg、1.75g/kg)均能降低高尿酸血症大鼠血清UA水平;葛根醇提剂量组(3.50g/kg、1.75g/kg)均能降低高尿酸血症大鼠血清TG水平。②造模第15天,葛根水提液(5.25g/kg、1.75g/kg)均能降低血清UA、TG水平;葛根醇提液(5.25g/kg、3.50g/kg、1.75g/kg)均能降低血清UA水平;葛根醇提液(3.50g/kg、1.75g/kg)均能降低血清TG水平。③造模第25天,葛根水提液、醇提液(5.25g/kg、3.50g/kg、1.75g/kg)均能降低高尿酸血症大鼠血清UA、TG水平、血浆粘度(PV);葛根醇提液(5.25g/kg、3.50g/kg、1.75g/kg)均能降低高、中、低切变率下的全血粘度(WBV);葛根水提液、醇提液(5.25g/kg)均能降低血清TC水平。[结论]葛根水提取物和醇提取物均具有降尿酸和调节血脂作用,其中醇提取物起效快,且能同时降低血浆粘度和全血粘度。 [Objective]To study the effects of Pueraria lobata extracts on modulating the levels of blood uric acid and blood lipid of hyperuricemia rats treated by purine-rich diet.[Methods]Ninety SD rats were randomly divided into normal group,model group,P.lobata water extract groups(5.25 g/kg,3.25 g/kg and 1.75 g/kg),P.lobata ethanol extract groups(5.25 g/kg,3.25 g/kg and 1.75 g/kg)and positive control group in average.The rats were fed with purine-rich diet consecutively for 25-days to establish the hyperuricemia model.The treatment groups were simultaneously orally administered with Pueraria lobata extracts for 25 days.During this trial,the general behavior and body weight were daily observed.After the administration for 8 days and 15 days and 25 days,the rat serum uric acid(UA),total cholesterol(TC)and triglyceride(TG)levels were separately detected using the orbit blood.1h after the last administration,the UA,TC,TG and hemarheological parameters were detected.[Results](1)After 8 day administration,the P.lobata ethanol extracts(5.25g/kg,3.50g/kg and 1.75g/kg)can reduce the UA level,and two of them(3.50g/kg and 1.75g/kg)can reduce the TG level.(2)After 15 day administration,the P.lobata water extracts(5.25g/kg,3.50g/kg and 1.75g/kg)can reduce the UA level and two of them(5.25g/kg and 1.75g/kg)can also reduce the TG level,while two of its ethanol extracts(3.50g/kg and 1.75g/kg)can reduce the TG level.(3)After 25 day administration,both of all water extracts and ethanol extracts of P.lobata can reduce the plasma viscosity,UA and TG levels,the ethanol extracts(5.25g/kg 3.50g/kg and 1.75g/kg)can reduce the whole blood viscosity(WBV),and the water extract and ethanol extract at the concentration of 5.25g/kg can reduce the TC level.[Conclusion]P.lobata extracts especially the ethanol ones have a therapeutic effect on hyperuricemia,with an activity of reducing plasma viscosity and blood lipid.
