摘要
目的:探讨低分子肝素对脂多糖诱导的人颊黏膜成纤维细胞分泌IL-8的影响。方法:原代培养正常人颊黏膜成纤维细胞,脂多糖刺激诱发炎症。不同浓度低分子肝素作用于炎症模型。采用ELISA方法检测细胞上清液中IL-8的变化。结果:脂多糖刺激后人颊黏膜成纤维细胞IL-8的表达高于正常对照组3 h(410.55 pg/mL与221.88pg/mL,P<0.05);低分子肝素组(1 U/mL)IL-8分泌量在6 h(552.85 pg/mL与885.81 pg/mL,P<0.05)、12 h(532.94 pg/mL与891.24 pg/mL,P<0.05)、24 h(405.96 pg/mL与887.88 pg/mL,P<0.05)较炎症组显著降低。结论:低分子肝素能显著抑制脂多糖刺激后人颊黏膜成纤维细胞IL-8的释放。
Objective:to explore the effects of low molecular heparin on IL-8 release from human buccal mucosa fibroblasts stimulated with lipopolysaccharide(LPS).Method:To primarily culture HBMFs.HBMFs excited by LPS,Different concentrations of low molecular heparin effects on inflammation in the model.ELISA method was used to measure IL-8 released into the culture medium from HBMPs in the different group s.Result:LPS increased IL-8 release from HBMFs at all the time points(P 0.05).LH inhibited the releases dose dependently(P 0.05).Conclusion:LMWH may inhibit the secretion of IL-8 of HBMFs stimulated with LPS.
