摘要
目的:探讨siRNA沉默NADPH氧化酶1(Nox1)基因表达对胃癌细胞的生长抑制作用。方法:合成Nox1 siRNA,采用实时定量PCR和Western blotting观察Nox1 siRNA转染后胃癌SGC-7901细胞Nox1 mRNA及相应蛋白表达的变化;用CCK-8细胞增殖实验、流式细胞检测技术分别检测胃癌SGC-7901细胞增殖及凋亡的变化。结果:转染Nox1 siRNA后的胃癌SGC-7901细胞Nox1 mRNA及蛋白表达明显受抑制。与对照组相比,干扰组SGC-7901细胞生长明显变缓(P<0.05),细胞凋亡率明显增高(P<0.01)。结论:Nox1 siRNA可明显下调靶基因Nox1的表达,在体外可抑制胃癌SGC-7901细胞的生长并促进其凋亡。
AIM: To investigate the inhibitory effect of synthetic NADPH oxidase 1(Nox1) short interfering RNA(siRNA) on the growth of human gastric cancer SGC-7901 cell line.METHODS: The siRNA targeting Nox1 mRNA was transfected into SGC-7901 cells and Nox1 expression was determined by real-time PCR and Western blotting.Cell proliferation and apoptosis were examined by the methods of CCK-8 and flow cytometry,respectively.RESULTS: Nox1 siRNA significantly inhibited the expression of Nox1 in gastric cancer cells at both mRNA and protein levels.The apoptotic rate of SGC-7901 cells transfected with siRNA was significantly higher than that of the control cells(P0.01).The proliferation of SGC-7901 cells was also significantly suppressed in vitro(P0.05) by siRNA transfection.CONCLUSION: Direct inhibition of Nox1 expression by siRNA significantly suppresses the proliferation of SGC-7901 cells in vitro,indicating a new approach of gene therapy for gastric cancer.
