摘要
目的研究抵抗素对人子宫内膜癌细胞系Ishikawa增殖、黏附及侵袭的影响。方法采用MTT法检测细胞的增殖活性及细胞黏附能力,实时定量PCR方法检测MMP-2、MMP-9及TIMP-1、TIMP-2的表达。结果10、50、100和150 ng/ml的抵抗素均能促进Ishikawa细胞的生长,并有时间和剂量依赖性,以100 ng/ml作用48 h的增殖作用最为明显,与对照组相比差异均有统计学意义(P<0.01);能增强细胞黏附能力,4个浓度的细胞黏附率分别为67%、69%、75%和98%,与对照组0%的黏附率比较差异有统计学意义(P<0.01);能促进MMP-2、MMP-9的表达,使MMPs/TIMPs的表达失衡,增强细胞侵袭能力。结论抵抗素可以促进子宫内膜癌细胞系Ishikawa的增殖,增强细胞的黏附及侵袭能力。
Objective To investigate the effect s of resistin on cell growth,adhesion and invasion of human endometrial carcinoma Ishikawa cells.Methods MTT assay was used to assess the proliferation and adhesion effects of resistin;and Real-Time PCR was adopted to detect the expressions of MMP-2,MMP-9,TIMP-1 and TIMP-2.Results Resistin at concentrations of 10,50,100 and 150 ng/ml promoted the proliferation of endometrial carcinoma cells in a time and dose-dependent manner(P0.01);the effect of resistin at 100 ng/ml for 48 h was most prominent.Resistin at a range of 10-150ng/ml enhanced the adhesive ability of Ishkawa cells,with the adherence rates of 67%,69%,75%and 98%,respectively(P0.01).Resistin enhanced the expressions of MMP-2, MMP-9,and induced the unbalance of MMPs/TIMPs ratio.Conclusion Resistin can promote the proliferation of endometrial carcinoma Ishikawa cells,and enhance its adhesive and invasive ability.
