鸡胚地龙膏活性肽抗氧化作用的研究

Study on the Antioxidant Effect of Active Peptide Extract from Jipeidilong Ointment

目的探讨鸡胚地龙膏活性肽提取物抗氧化作用。方法测定鸡胚地龙膏活性肽提取物在邻苯三酚自氧化体系中对超氧阴离子自由基、Fenton反应体系中对羟基自由基的清除作用。通过对新西兰大白兔急性软组织损伤模型测定其血液和组织抗氧化酶活性及丙二醛(MDA)含量,检测鸡胚地龙膏活性肽提取物在新西兰大白兔体内的抗氧化能力。结果体外试验研究证实,鸡胚地龙膏活性肽提取物能有效清除超氧阴离子自由基和羟基自由基的作用。体内试验中,鸡胚地龙膏高剂量治疗组血液及组织液中的SOD、CAT、GSH-PX活性水平分别与正常对照组、模型组、阳性药物组及鸡胚地龙膏低剂量组比较,差异有统计学意义(P<0.01);而血液及组织液中的MDA含量水平明显低于其他4组(P<0.01)。结论鸡胚地龙膏活性肽提取物具有较强的清除活性氧及抗氧化作用,可能是鸡胚地龙膏治疗急性软组织损伤的机制之一。

Objective To investigate the antioxidant effect of active peptide extract from jipeidilong ointment.Methods Pyrogallol autoxidation system and Fenton reaction system were adopted to detect the scavenging capacity against superoxide anion radical and hydroxyl radical.The model of acute soft tissue injury in Zealand white rabbits were established to detect the antioxidase activities and malondialdehyde（MDA） content in blood and tissue.Results The test in vitro proved that active peptide extract from jipeidilong ointment had good effect of scavenging superoxide anion radical and hydroxyl radical.The test in vivo showed that SOD,CAT,GSH-PX activities in blood and tissue of high-dose jipeidilong ointment group were higher than those in normal control group,model group,positive medicine group,and low-dose jipeidilong ointment group,and differences were statistically significant（P0.01）;the MDA content was significantly lower than that in other four groups（P0.01）.Conclusion The active peptide extract from jipeidilong ointment has stronger effect of scavenging active oxygen and antioxidant effect,can effectively reduce lipid peroxidation and scavenge active oxygen free radicals,which may be one of the mechanisms for the treatment of acute soft tissue injury with jipeidilong ointment.
