摘要
目的观察沥青烟对小鼠生殖系统损伤的影响,并探讨其发生机制。方法用60只雄性小鼠建立亚急性实验模型,吸入不同浓度、不同时间沥青烟。采用伊红染色观察精子畸形,放射免疫法测定睾酮水平,光镜下观察睾丸组织病理学变化。结果不同浓度沥青烟染毒2、4和8周,小鼠精子畸形率明显增加,畸形精子均以无定形和无钩为主;小鼠血清中睾酮含量降低,2和4周差异有统计学意义(P<0.05),8周随着染毒浓度增加,睾酮含量呈逐渐下降趋势;睾丸病理观察可见染毒组生精细胞层数减少,精子染色变淡,精细胞有不同程度变性坏死,曲细精管间质呈不同程度水肿。结论沥青烟对雄性小鼠具有亚急性生殖毒性。
Objective To observe the effects of bitumen fume on reproductive toxicity of male mice and probe into its mechanism. Methods 60 male mice were forced to inhale bitumen fume at different exposure levels with different time periods to establish sub-acute models. Deformity of sperm stained with eosin was observed. Testosterone of serum was measured by radioimmunoassay and pathological changes of testis were observed under microscope. Results When exposed to different fume concentrations for 2, 4 and 8 weeks, the sperm deformity rates increased significantly mainly showing amorphous and without hook. The contents of serum testosterone lowered. The difference of results between 2 and 4 weeks was significant(P〈0.05) After exposed for 8 weeks, as the exposure levels increased, the contents of testosterone showed gradual decreasing trend. The pathological changes of testis showed the reduced layers of spermatogenic cell, weakly stained sperms,sperm necrosis and the swollen mesenchyme of convoluted tubule in certain extent. Conclusions Bitumen fume could induce the sub-acute toxicity of reproductive system in male mice.
