摘要
目的:检测胆囊成纤维细胞生长因子19(fibroblast growth factor 19,FGF19)的表达,并探讨正常人空腹及餐后血浆FGF19的含量与胆囊容积变化的相关性。方法:采用定量PCR和免疫组化检测胆囊黏膜FGF19 mRNA和蛋白质的表达。采用酶联免疫吸附法测定8例健康志愿者(男女各4例)空腹及脂肪餐后1~4 h血浆FGF19含量,B超检查相应时段的胆囊容积。结果:胆囊具有较高的FGF19 mRNA表达量。FGF19蛋白在胆囊黏膜上皮细胞表达。脂肪餐后胆囊收缩,在餐后2 h达到最高峰。脂肪餐后1 h,女性血浆FGF19含量略有降低,之后逐渐上升,于3 h达到最高峰;这种波动稍早于胆囊容积的变化;而男性没有类似的波动。结论:人胆囊黏膜上皮表达FGF19。脂肪餐后,血浆FGF19和胆囊容积的变化可能具有一定的相关性。
Objective Fibroblast growth factor 19(FGF19) is involved physiologically in the refilling of gallbladder.To measure the expression of FGF19 in gallbladder tissue and to evaluate the relationship of the plasma FGF19 level and the changes of gallbladder volume.Methods mRNA level of FGF19 in gallbladder tissues were quantified by real-time PCR.Localization of FGF19 protein in gallbladder tissues was detected by immunohistochemistry.FGF19 in plasma was assayed by ELISA during fasting and 1~4 hour after fat-diet obtained from 4 female and 4 male healthy volunteers.At each time point,the gallbladder volume was measured using B-type ultrasonic scanning.Results mRNA of FGF19 was detected in gallbladder tissue.Immunohistochemistry showed that FGF19 protein was present in the epithelium of gallbladder.The gallbladder contracted after fat-diet and reached the maximum degree 2-hour after the diet in both sexes.In females,plasma FGF19 decreased 1-hour after the fat-diet,rose afterwards,and reached the peak 3-hour after the fat-diet.This change was earlier than the refilling of the gallbladder.However,similar findings were not observed in males.Conclusions Our study shows that FGF19 is highly expressed in the human gallbladder epithelium.Plasma FGF19 may be related with gallbladder refilling after fat-diet.
