摘要
目的探讨兔VX2肾移植瘤TACE治疗前后ADC值与细胞密度变化的对应关系,明确DWI在肾脏肿瘤治疗后随访的生物学基础。方法建立兔VX2肾移植瘤动物模型,并随机分为对照组和TACE组。对照组经肾动脉注射生理盐水10ml,TACE组注射超液态碘油2 mg/kg和阿霉素2 mg/kg。TACE术前、术后7 d行常规MRI及扩散加权成像检查,记录治疗前后肿瘤磁共振表现特征及表现扩散系数(ADC值)。细胞密度根据HE染色图片进行分析。结果 TACE组和对照组肿瘤细胞密度分别为(24.05±8.13)%和(36.03±3.13)%(P=0.022)。TACE术后ADC值为(1.321±0.125)×10-3mm2/s,高于对照组[(1.146±0.133)×10-3mm2/s](P=0.004)。两组肿瘤细胞密度均与ADC值呈负相关(r对照组=-0.697,P=0.025;rTACE=-0.615,P=0.033)。结论兔肾VX2移植瘤TACE治疗后ADC值变化取决于肿瘤组织细胞密度的变化,DWI能反映肿瘤治疗前后组织微观结构的变化。
Objective To explore the biological basis of DWI in the evaluation of the effect of transcatheter arterial chemoembolization(TACE)on VX2 tumor implanted in rabbit kidney by analyzing the relationship between apparent diffusion coefficien(ADC)value and cell density of tumor before and after the treatment.Methods The rabbits bearing VX2 were built and divided into two groups randomly,the control group(n=10)and TACE group(n=12).The normal saline was administered via the kidney artery in the control group.While lipiodol(0.2 ml/kg)and ADM(2 mg/kg) were used in TACE group.Conventional MRI and DWI were performed before and after the treatment.The imaging features and ADC value were recorded.Cell density of tumor was analyzed by HE staining.Results The cell density of TACE group and the control group were(24.05±8.13)% and(36.03±3.13)%(P=0.022).The ADC value of TACE group was(1.321±0.125)×10^-3mm^2/s,which was higher than that of the control group[(1.146±0.133)×10^-3mm^2/s](P=0.004).The ADC value was correlated with cell density negatively before and after treatment(rbefore TACE=-0.697,P= 0.025;rafter TACE=-0.615,P=0.033).Conclusion DWI could reflect the histology change of the VX2 implanted tumor in rabbit kidney before and after the treatment,which has the potential to evaluate the effect of TACE.
