摘要
目的通过观察同型半胱氨酸对人血管平滑肌细胞死亡受体Fas、TNFR1mRNA表达的研究,探索同型半胱氨酸诱导血管平滑肌细胞凋亡的分子机制。方法应用RT-PCR技术对死亡受体Fas、TNFR1mRNA的表达进行观察。结果 RT-PCR技术检测结果发现同型半胱氨酸使血管平滑肌细胞Fas、TNFR1mRNA的表达呈浓度和时间依赖性上调(P<0.05)。结论同型半胱氨酸诱导人血管平滑肌细胞死亡受体Fas、TNFR1 mRNA表达增加,这可能是同型半胱氨酸诱导血管平滑肌细胞凋亡的机制之一。
Objective To investigate the effects of Homocysteine(Hcy)on the expression of Fas and TNFR1 mRNA of human vascular smooth muscle cells(HVSMCs)cultured in vitro.Methods The expressions of Fas and TNFR1 mRNA in human vascular smooth muscle cells were detected by the way of reverse-transcriptase polymerase chain reaction(RT-PCR).Results the ratio of the absorbency of Fas、TNFR1mRNA/β-actin of HVSMCs incubated with 500 μmol/L Hcy for 0 h、6 h、12 h、24 h、48 h and incubated with 0、100、200、500、1000 μmol/L Hcy for 24 h was upregulated in a concentration and time-dependent manner(P0.05).Conclusion Homocysteine can enhance the expressions of death receptors Fas、TNFR1mRNA in human vascular smooth muscle cells,the two receptors-mediated signaling pathways may be one of the mechanisms of apoptosis in atherosclerosis of vascular smooth muscle cells induced by homocysteine.
