HPLC法同时测定丹参当归复方制剂中丹酚酸B和阿魏酸的含量 被引量:4

摘要 目的建立一种简便的能够同时定性定量检测丹参当归复方制剂中丹酚酸B和阿魏酸的方法。方法 HPLC法,色谱万能柱ODS-C18(250 mm×4.6 mm,5μm)柱,流动相:甲醇/乙腈/甲酸/水,检测波长为286 nm。流速0.8 ml/min。结果丹酚酸浓度在5.40~43.25μg/ml,阿魏酸浓度在3.37~27.00μg/ml之间线性关系良好。线性方程分别为:丹酚酸By=0.971 1x-2.215 4,R2=0.999 6,阿魏酸y=0.415 9x-0.450 7,R2=0.999 8(y:阿魏酸/丹酚酸B的浓度μg/ml,x:峰面积万)。平均回收率为98.4%,98.7%,RSD分别为3.39%,1.49%,n=6。结论 HPLC在检测丹酚酸B和阿魏酸时有较好的专属性,灵敏度高,重现性好,且样品处理简单容易操作,可以作为丹参和当归复方制剂的质量检测方法。 Objective To establish an expedited and simplified method for the contemporary detection of salvianolic acid B and ferulic acid in quality and quantity.In these herbage stuffs phenolic acids are their mainly availability compounds,such as salvianolic acid B and ferulic acid are the representatives.Methods An external standard method by HPLC with ODS-C18(250 mm×4.6 mm,5 μm)column.The mobile phase was methanol/acetonitrile/eormic acid/water and flow rate was 0.8 ml/min.The UV detector wavelength was set at 286nm.Results The linear range of salvianolic acid B was 5.40-43.25 μg/ml,and ferulic acid was 3.37-27.00 μg/ml.The linear equations: Y=0.971 1X-2.215 4,R2=0.999 6,and Y=0.415 9X-0.450 7,R2=0.999 8.The average recycle rate were 98.4%,98.7%,and RSD 3.39%,1.49%(n=6) respectively.Conclusion The above-mentioned method is good attributive to detect salvianolic acid B and ferulic acid,has high sensitivity and well repetition.And sample can be easily and ordinaryly treated.So this method can be adoptable for quantitative analysis of salvianolic acid B and ferulic acid in compound preparations of danshen and danggui.
