碳酸钙固定化猪胰脂肪酶的制备及活性研究 被引量:3

摘要 本文利用市售普通碳酸钙(general calcium carbonate,GCC)作为猪胰脂肪酶(PPL)固定化的载体材料,研究了不同固定化条件对所制备的固定化酶活性的影响。结果显示PPL固定化于GCC上的最佳制备工艺为:将20 mL酶液与1.50 g GCC放入置于摇床内的三角瓶中,摇床温度设为40℃,摇床转速设为180 rpm,固定化时间为2 h,所得固定化酶的最大活性可达到141Us。 The purchased CaCO3 was used as the supports for the immobilization of porcine pancreatic lipase(PPL),and the influences of various experimental parameters,such as the mass ratio of PPL and CaCO3,incubation temperature,incubation time and the rotating speed of shaker on the activity of PPL were investigated in detailed.The optimal immobilization conditions for PPL could be described as follows:1.50 g CaCO3 was matched with 20 mL PPL solution,the incubation temperature was 40℃,the rotating speed of shaker was 180 rpm,the incubation time was fixed at 2 h.After this procession,the activity of prepared immobilized PPL can be achieved its maximum value of 141Us.
