TSHR和RASSF1A基因甲基化与乳头状甲状腺癌的相关性 被引量:2

摘要 目的:研究乳头状甲状腺癌TSHR和RASSF1A基因启动子甲基化的状况,分析两种抑癌基因甲基化与临床病理资料间的关系及两种抑癌基因甲基化的相关性,探讨PTC的发生机制。方法:提取50例PTC组织和32例对照组织DNA,使用甲基化PCR检测两种基因启动子区甲基化的情况;对两种基因甲基化和未甲基化的组织测序;采用SPSS13.0软件分析两种抑癌基因甲基化之间的关系及两种抑癌基因启动子甲基化和主要的临床病理参数的关系。结果:50例PTC中,发现34例TSHR基因、30例RASSF1A基因启动子发生甲基化;32例对照组中,7例TSHR基因、10例RASSF1A基因启动子发生了甲基化;PTC组TSHR基因和RASSF1A基因启动子甲基化率均显著高于对照组,差异有统计学意义。DNA测序证实,两种抑癌基因发生甲基化的其CpG岛的碱基仍为CG,未发生甲基化的碱基由CG变为TG。计算TSHR和RASSF1A基因甲基化间的相关系数r=0.490,提示PTC组两个抑癌基因甲基化之间存在显著相关性。RASSF1A和TSHR基因甲基化与患者的年龄、性别、临床分期均未见相关性;RASSF1A基因甲基化与患者是否伴有淋巴结转移无关,TSHR基因甲基化与患者是否伴有淋巴结转移相关。结论:两种抑癌基因启动子甲基化与PTC的发生有关,且可能在甲状腺肿瘤的早期阶段既已存在;TSHR基因甲基化在甲状腺癌的恶性进展中可能发挥一定作用。
