摘要
目的观察增损甘露消毒丹对肝纤维化大鼠肝组织PDGF-BB、CTGF的影响,探讨其抗肝纤维化的作用机制。方法采用四氯化碳(CCL4)皮下注射和乙醇灌胃等制备肝纤维化大鼠模型,设空白组、病理模型组、增损甘露消毒丹小、中、大剂量组和复方鳖甲软肝片组。治疗结束后肝组织作HE染色观察肝组织纤维化程度,免疫组织化学方法(SP法)检测肝组织PDGF-BB、CTGF的表达。结果与模型组比较,增损甘露消毒丹大、中剂量组肝损害明显好转,肝细胞变性坏死程度较轻,结缔组织增生不明显;大鼠肝组织PDGF-BB、CTGF的含量明显降低,差异有显著性意义(P<0.05)。结论增损甘露消毒丹能够改善肝纤维化程度,拮抗肝纤维化大鼠肝组织中PDGF-BB、CTGF的表达,从而发挥抗肝纤维化的作用。
Objective To observe the effects of Zengsun Ganlu Xiaodu pills on the liver tissue PDGF-BB and CTGF of rats with hepatic fibrosis,and to probe into its mechanism of anti hepatic fibrosis.Methods Hepatic fibrosis rats model was made with subcutaneous CCL4 injection and intragastric ethanol administration.The rats were divided into normal control group,pathologic model group,the low,medium and high doses of Zengsun Ganlu Xiaodu pills groups,and compound Biejia Ruangan pills group.At the end of the treatment,liver tissues were observed with HE staining to observe the extent of fibrosis.Immunohistochemical method(SP) was used to detect the expression of PDGF-BB and CTGF.Results Compared with pathologic model group,the liver damage of rats in high and medium Zengsun Ganlu Xiaodu pills groups got better obviously,the extent of liver cells' degeneration and necrosis was less severe,the hyperplasia of connective tissue was not distinct,and the content of PDGF-BB and CTGF in rats' liver tissues was markedly reduced.The difference was significant(P〈0.05).Conclusion Zengsun Ganlu Xiaodu pills have the effect of anti hepatic fibrosis by lessening the extent of hepatic fibrosis,and resisting the expression of PDGF-BB and CTGF in the liver tissue of rats with hepatic fibrosis.
