穿心莲生殖毒性的时效关系研究 被引量:3

摘要 研究穿心莲对雄性小鼠精子、酶活力及睾丸组织结构的影响,并探讨时效关系.实验分为对照组、穿心莲提取物组和穿心莲内酯组,分别给予蒸馏水、4.67 g/kg穿心莲提取物溶液和315 mg/kg穿心莲内酯溶液,经口灌胃,每天1次,给药持续77 d(11周).分别在给药的第21、35、49、63、77 d剖杀受试小鼠,称量体重和主要脏器质量,观察精子数和精子形态的变化,测定睾丸中乳酸脱氢酶(LDH)及碱性磷酸酶(AKP)的活力并制备睾丸及附睾病理切片.与对照组相比,受试小鼠的睾丸及前列腺的绝对质量和相对质量均显著降低(P<0.05);小鼠精子数明显低于对照组,精子畸形率高于对照组(P<0.05);随着给药时间的增加,小鼠睾丸匀浆中LDH、AKP活力明显下降(P<0.05);受试小鼠的睾丸中曲细精管不同程度萎缩,生精细胞数量降低;附睾可见附睾管管腔上皮增厚,精子大量减少.穿心莲提取物和穿心莲内酯对雄性小鼠具有生殖毒性作用,且存在一定的时效关系. Studied the effects of andrographis on male mice sperm、enzyme activity and organizational structure,and to discuss its time-effect relationship.The experiment were divided into control group,andrographolide extraction and andrographolide group were given distilled water,4.67 g/kg andrographolide solution and 315 mg/kg andrographolide solution,by oral gavage and administrate 1 time each day,continued for 77 d(11 weeks).Tested mice were killed on the 21,35,49,63,77 d of administration,the weights of tested mice and major organ were measured,observed the changes of sperm count and sperm morphology,the testis and epididymis pathological were prepared,at the same time the activities of LDH and AKP of testis were also measured.Compared with the control group,the absolute and relative weights of testis and prostate of tested mice decreased significantly(P0.05);the treated groups sperm number were significantly lower than control,sperm malformation rate were higher than control group(P0.05);with the increases of exposure time,the activities of LDH and AKP in the testis of tested mice decreased significantly(P0.05).the testis of tested mice appear different degrees of testicular semiferous tubule atrophy and decrease in the number of spermatogenic cells.The epididymis appear different degrees of luminal epithelium thicken and decrease in the number of sperm.andrographolide extraction and andrographolide have reproductive toxicity effects on male mice,and its toxicity exist has certain time-effect relationship.
