摘要 研究β-葡聚糖酶对β-1,3-葡聚糖酶解的最佳条件.采用DNS显色法分别测定底物质量浓度、酶质量浓度、酶解的温度、pH值对酶解产物还原性糖质量分数的影响;在单因素基础上做正交优化试验,再对酶解时间进行考察.结果表明,最佳酶解条件为糖质量浓度1.4 mg/mL,酶质量浓度14 mg/mL,温度55℃,反应pH值为5.5,反应时间10 min,在此条件下酶解产物还原性糖质量分数最大,为95.30%. This paper studied on enzymolysis of β-1,3-glucan in the optimum of enzyme condition by β-glucanase.Effects of substrate concentration,enzyme amount,reaction temperature and reaction pH on yield of reducing sugar content were studied.On this basis,orthogonal were designed to investigate the enzymatic hydrolysis of time.The results showed that the optimum substrate was 1.4 mg/mL,enzymatic hydrolysis was 14 mg/mL,and temperature was 55 ℃,pH 5.5 for 10 minutes.In the condition,a maximum of reducing sugar hydrolysates yield was 95.30%.
