绿脓杆菌耐药性基因重组蛋白诱导时间的优化

摘要 绿脓杆菌是一类人与动物的条件致病菌,极易产生耐药性,且对极大部分抗生素均不敏感。因而寻找一种有效靶标获得针对性强的新型抑制剂显得尤为迫切。烯酯酰-ACP还原酶是脂肪酸延伸循环中的一个关键酶,所以它是一种较为理想同时又具有一定选择性的抗菌药物靶位。本实验通过绿脓杆菌的烯酯酰-ACP还原酶IV阳性克隆转染、诱导后所提纯的蛋白酶能在外源宿主细胞内以可溶性蛋白的形式表达,从而优化蛋白诱导表达时间,使绿脓杆菌烯酯酰-ACP还原酶IV重组蛋白酶达到较高的活性表达。为建立以烯酯酰-ACP还原酶IV为靶标的新型抗生素筛选模型奠定了基础。
