摘要
目的探讨三氯乙烯(TCE)药疹样皮炎患者外周血免疫相关基因Foxp3、GATA3、CTLA4、T—bet的mRNA表达。方法选取TCE药疹样皮炎患者8例为病例组,8例健康人为对照组,抽取两组对象的外周血,采用实时荧光定量PCR(real—timequantitativePCR)技术检测外周血免疫相关基因Foxp3、GATA3、CTLA4、T—bet的mRNA的表达水平。结果与对照组比较,病例组Foxp3、GATA3、CT—LA4基因mRNA表达水平分别升高115%、97%和241%,差异有统计学意义(P〈0.01);与对照组比较,病例组T—bet基冈mRNA表达水平下降47%,差异有统计学意义(P〈O.01)。结论TCE药疹样皮炎的部分免疫相关基斟表达水平发生明显改变,这些基因可能在TCE引起的变态反应中发挥重要作用。
Objective To study mRNA expression of immune-related genes (Foxp3, GATA3, CTLA4 and T-bet) in peripheral blood of the patients with allergic dermatitis induced by trichloroethylene (TCE). Methods The peripheral blood samples were collected from 8 healthy workers (control group) and 8 patients with allergic dermatitis induced by TCE (case group). Real-time quantitative PCR was applied to detect mRNA expression of immune-related genes (Foxp3, GATA3, CTLA4, T-bet). Results The mRNA expression levels of Foxp3, GATA3 and CTLA4 genes increased by 115%, 97% and 241% in case group, as compared with con-trol group (P〈0.01). The mRNA expression level of T-bet gene decreased by 47% in case group, as compared with control group (P〈0.01). Conclusion The mRNA expression levels of some immune-related genes changed in patients with allergic dermatitis induced by TCE, those genes may play an important role in TCE-induced allergy.
