摘要
目的检测12-O-十四烷酰佛波醋酸酯-13(TPA)处理C57BL/6(B6)小鼠背部皮肤后TCF3表达的变化情况。方法采用免疫荧光染色、RT-PCR和Western blot等技术,检测TPA和丙酮分别处理7周大小的B6鼠背部皮肤1、2和4周后TCF3的表达情况。结果与丙酮处理组相比,TPA处理组中B6鼠背部皮肤明显增厚,并且在TPA处理4周后毛囊由静止期形态转变为生长期中期形态。TPA处理1周和2周后,TCF3的蛋白和mRNA表达上调,主要核表达在毛囊bulge以及次级毛芽处。处理4周后TCF3的蛋白和mRNA表达上调,主要定位在毛囊bulge及其下段的毛囊外根鞘的毛囊干细胞内;而在丙酮处理组可见TCF3一直表达在毛囊bulge处。结论 TPA能在促进毛囊干细胞异常活化的同时,促进TCF3的表达。
Objective To study TCF3 expression in back skin of C57BL/6(B6) mice in treatment of tetradecanoylphorbol acetate(TPA).Methods Seven-week old B6 mice were treated with TPA(TPA group) or acetone(acetone group) on their back skins for 1,2 and 4 weeks,respectively.Immunofluorescence,RT-PCR and Western blotting were applied to examine TCF3 expression in hair follicles of mice.Results Compared with the acetone group,the back skins of the TPA group were thicker,and the resting hair follicles transformed into growing hair follicles after the treatment of TPA for 4 weeks.After the treatment of TPA for 1 and 2 weeks,the protein and mRNA level of TCF3 was up-regulated and TCF3 was mainly expressed in the nuclei of hair follicle bulge and secondary hair germ.After the treatment of TPA for 4 weeks,TCF3 was upregulated in the protein and mRNA level and was mainly expressed in hair follicle bulge and hair follicle stem cells in the out root sheath.However,TCF3 of the acetone group was expressed in hair follicle bulge and secondary hair germ all the time.Conclusion TPA can promote abnormal activation of hair follicle stem cells and meanwhile induce up-regulation of TCF3.
