摘要
目的研究多棘海星乙醇提取物对人肿瘤细胞体外生长的影响,为进一步筛选抗肿瘤活性成分提供重要的科学依据。方法用硅胶柱层析法对多棘海星乙醇提取物进行组分分离,采用MTT法观察各个组分对体外培养的人乳腺癌细胞MDA-MB-231、人宫颈癌Hela细胞、鼻咽癌CNE-1细胞等肿瘤细胞株增殖的抑制作用。结果通过硅胶柱层析从多棘海星乙醇提取物中分离得到8个主要组分(Ⅰ~Ⅷ)。MTT结果显示多棘海星乙醇提取物中的4个组分对肿瘤细胞增殖具有抑制作用,其中组分Ⅳ和Ⅷ对MDA-MB-231、Hela和CNE-1细胞的增殖具有非常显著的抑制作用(P<0.01),组分Ⅴ对Hela细胞增殖具有一定的抑制作用(P<0.05),组分Ⅶ则对Hela细胞的增殖具有抑制作用(P<0.01),而组分Ⅰ、Ⅱ、Ⅲ和Ⅵ则对上述3种细胞增殖均无抑制作用(P>0.05)。结论多棘海星乙醇提取物对肿瘤细胞的增殖具有抑制作用,但不同组分对不同的肿瘤细胞株的增殖表现出不同的抑制作用,组分Ⅳ和Ⅷ可能是其主要的抗肿瘤活性成分存在部位。
Objective To investigate the antitumor activity of the ethanol extracts of starfish Asterias amurensis in vitro. Methods Silica gel column chromatography was used to separate the fractions of the ethanol extracts of starfish Asterias amurensis. In vitro, the inhibitory effects of ethanol extracts of starfish Asterias amurensis on proliferation of human breast carcinoma cell (MDA-MB-231), cervical cancer cell line (Hela Cell) and nasopharyngeal carcinoma cell line(CNE-1) were measured by MTT colormetric assay, Results The ethanol extracts from starfish were separated in silica gel column to obtain 8 fractions (I-VIII), 4 of which could inhibit carcinoma cells proliferation. The fraction IV and VIII could significantly inhibit the proliferation of MDA- MB-231, Hela and CNE-1 cells (P〈0.01), fraction V also could inhibit the proliferation of Hela cell (P〈0.05), and fraction VII showed the inhibitory effects on proliferation of Hela (P〈0.01), while the other 4 fractions of I, II, III and VI did not show the inhibitory effects on carcinoma cell proliferation(P〉0.05), Conclusion The ethanol extracts of starfish Asterias amurensis possess significant inhibitory effect on carcinoma cells, and the fractions IV and VIII may be the major fraction contains antitumor active substance.
