摘要
在硅胶柱层析获得暗孢节菱孢菌(Arthrinium phaeospermum)非蛋白毒素基础上,以浸渍法进行生测和叶圆片法分析不同浓度毒素对撑×绿杂交竹(Bambusa pervariabilis×Dendrocalamopisis grandis hybrid bamboo)叶组织细胞膜透性、可溶性蛋白、可溶性糖、丙二醛(MDA)、游离脯氨酸(Pro)含量及过氧化物酶(POD)、过氧化物歧化酶(SOD)和苯丙氨酸解氨酶(PAL)活性的影响。结果表明:暗孢节菱孢菌毒素可使杂交竹呈现典型症状,且使细胞膜透性和MDA含量显著提高。在较低毒素浓度范围内,毒素与杂交竹体内的游离脯氨酸含量、防御酶活性呈正相关,与可溶性糖负相关;而当浓度较高时,毒素与游离脯氨酸含量以及防御酶活性呈负相关,这说明高浓度毒素能使杂交竹机体组织完全破坏,从而证明了毒素在暗孢节菱孢菌对杂交竹致病过程中起重要作用。
According to the nonprotein toxin isolated by silica gel column chromatography,bioassay was tested by impregnation method in hybrid bamboo.Moreover,effects of different concentrations of toxin on the relative permeability of cell membrane,the contents of soluble protein,soluble sugar,malondialdehyde(MDA),free proline(Pro) and the activities of superoxide dismutase(SOD),peroxidase(POD),pheny-lalanine amonnialyase(PAL) were analyzed by the leaf disk method.The results showed that the typical symptoms were indicated in the toxin treatments.The relative permeability of cell membrane and the content of MDA were significantly increased.When treated with the low concentrations of toxin,there was a positive correlation between the content of free Pro,the activities of enzymes and toxin.The correlation between the content of soluble sugar and the concentration of toxin was negative.However,when treated with the high concentrations of toxin,there was a negative correlation between the content of free Pro,the activities of enzymes and toxin.This demonstrated that high concentrations of toxin could destroy the tissues of hybrid bamboo and toxin played an important role in pathogenicity of A.phaeospermun.
