摘要
目的观察中药益神启窍方对弥漫性轴突损伤合并局灶性脑挫伤大鼠β-APP表达的影响及可能机制。方法采用弥漫性轴突损伤合并局灶性脑挫伤动物模型造模,取雄性SD大鼠36只,随机分为4组,正常组7只、假手术组7只、模型组12只、中药组12只,后2组再随机分为创伤后6h、12h、24h3组,每组4只。造模前1周中药组每天灌胃益神启窍方中药配方颗粒,模型组只灌胃等剂量的生理盐水,分别用HE染色和β-APP免疫组化染色,观察造模后6h、12h、24h大鼠脑组织病理学改变和β-APP的表达。结果假手术组与正常组β-APP表达差异无统计学意义(P>0.05);中药组6h、12h、24hβ-APP表达均低于模型组、有显著性差异(P<0.05),但仍高于假手术组,有显著性差异(P<0.05)。HE染色正常组、假手术组无异常;模型组可见脑实质水肿,血管扩张淤血,蛛网膜下腔出血,部分神经元肿胀和少量核固缩,中药组见脑实质水肿程度、血管扩张淤血轻微,见少量的蛛网膜下腔出血,部分神经元轻微肿胀和少量核固缩。结论中药益神启窍方能够降低损伤脑组织中β-APP的表达,这可能与其具有脑保护作用有关。
OBJECTIVE To observe the effects and possible mechanisms of Yishen Qiqiao Fang on the expression of β-APP in the brain of rats with diffuse axonal injury and focal cerebral contusion.METHODS Diffuse axonal injury and focal cerebral contusion animal model was adopted as a sample.36 male SD rats were randomly divided into four groups: 7 in normal group,7 in sham-operated group,12 in control group and 12 in Chinese drug group.The last two groups were randomly divided into three groups,namely post-traumatic 6 h,12 h and 24 h,four in each group.Yishen Qiqiao Granule was lavaged in Chinese drug group every day one week before modeling,while the same amount of physiological saline was only lavaged in model group.Both groups were stained with HE and β-APP immunohistochemistry respectively,and then cerebral histopathological changes and β-APP expressions of rats were observed 6h,12 h and 24 h after modeling.RESULTS There was no significant differences in sham-operated group and normal group with regard to β-APP expression(P0.05);The β-APP expressions of Chinese drug group 6 h,12 h,and 24 h were all lower than that of control group,indicating significant differences(P0.05),but they were still higher than that of sham-operated group,suggesting significant differences(in 12 h)(P0.05).HE staining was normal in normal group and sham-operated group;cerebral parenchyma edema,vascular congestion expansion,subarachnoid hemorrhage,a part of neuronal swelling and a small portion of nuclear condensation were presented in control group,whereas the mild cerebral parenchyma edema and vascular congestion expansion,a small amount of subarachnoid hemorrhage,a part of slight neuronal swelling and mild nuclear pyknosis were shown in Chinese drug group.CONCLUSION Yishen Qiqiao Fang can reduce β-APP expression in damaged brain tissues,which may be associated with its function of protecting the brain.
