基于反相液相色谱的人血中茶碱浓度临床检测与分析

摘要 目的:建立快速、简便、准确、重现性好的人血中茶碱的测定方法。方法:采用Hypersil ODS C18(4.6 mm×250 mm,5μm)为色谱柱,流动相为甲醇-水(25∶75),内标物为咖啡因,检测波长为274 nm,血清用甲醇沉淀蛋白后进行分析。结果:茶碱标准曲线回归方程为Y=0.053 9X+0.038,r=0.999 4(n=8),1~40μg/ml浓度范围线性关系良好。对36例患者作血清茶碱浓度监测,临床效果良好。结论:本测定方法为临床药师指导临床有效、安全的个体化药物的使用提供了保障。 Objective: To develop a rapid, simple, accurate and reproducible method for the determination of theophylline in human serum. Methods: The separation was performed on Hypersil ODS C18 column (4.6 mm×250 mm, 5 μm) with mo- bile phase consisted of methanol-water (25:75), and the wavelength for detection was 274 nm. The coffeine was used as an internal standard, and methanol was used to precipitate the serum protein. Results: The regression equation of theophylline was Y=0.053 9X+0.038, r=-0.999 4 (n=8). The calibration curves of theophylline were linear over the range of 1-40 μg/ml. The clinical application was good for 36 patients through of the detection of theophylline. Conclusion: The method sup- plies a reasonable guide for the drug use at individual levels.
