摘要
目的:探讨FHIT、PCNA在宫颈鳞状细胞癌及癌前病变中的表达规律,研究其在宫颈癌变中的作用和意义。方法:采用免疫组织化学(S-P法)检测FHIT、PCNA在18例正常宫颈组织、52例CIN和69例宫颈癌中的表达。结果:FHIT在正常宫颈组织、CINⅠ、CINⅡ、CINⅢ和宫颈鳞癌组织中阴性率分别为0%、9.1%、29.4%、41.7%、66.7%,正常组与宫颈癌组之间、CIN组与宫颈癌组之间、CINⅠ、Ⅱ组与CINⅢ组之间比较,差异均有显著性(P<0.05);随着细胞分化的降低,FHIT阴性率上升。PCNA在宫颈癌中的阳性率为84.1%,高于CIN和正常组(P<0.05),CIN和宫颈癌两组FHIT阴性者的PCNA阳性率明显高于FHIT正常表达者(P<0.01),FHIT与PCNA呈负相关关系。结论:FHIT、PCNA蛋白异常表达与宫颈癌的发生密切相关,FHIT缺失可作为判断CIN有可能发展为浸润癌的有用指标。CIN和宫颈鳞癌中的FHIT缺失常与PCNA过度表达同时存在,两者存在负相关关系。
Objective:To study the expression of FHIT and PCNA in cervical squamous Cell carcinoma(SCC) and cervical intraepithelial neoplasia(CIN) and to investigate the role and significance of them.Methods:The expression of FHIT protein and PCNA protein was determined using immunohistochemistry(S-P) in 18 cases of normal cervical squamous epithelium,52 cases of CIN and 69 cases of cervical SCC.Results:The rates for loss of expression of FHIT genes in the squamous epithelium of normal cervices,CIN Ⅰ,CIN Ⅱ,CIN Ⅲand invasive cervical SCC were 0%,9.1%,29.4%,41.7%,66.7%.The comparison between normal cervical squamous epithelium(NEC) and cervical SCC,the comparison between CIN and cervical SCC,the comparison between CIN Ⅰ、Ⅱ and CIN Ⅲ have statistic significance(P0.05).Along with the decreasing of cell differentiation,the negative rate of FHIT raised.The positive expression of PCNA in cervical SCC group was 84.1%,higher than that in CIN and normal groups(P0.01).The positive rate of PCNA in the depleted expression of FHIT group was significantly higher than that in FHIT normal expression group(P0.01).There is a negative correlation between FHIT And PCNA expression.Conclusion:The FHIT-depletion and PCNA over-expression are related with cervical carcinogenesis.The FHIT-depletion may be a clinically useful marker of CIN more likely to progress to invasive disease;Depleted expression of FHIT is freguently associated with PCNA over-epression in CIN and cervical SCC subjects.There is a negative correlation between FHIT And PCNA expression.
