摘要
利用发财蔓绿绒茎尖、茎段作为外植体,研究不同浓度6-BA和NAA对丛生芽诱导和增殖及壮苗和生根的影响。结果表明:不同外植体不定芽诱导率不同,茎尖诱导率最高,为91.50%,茎段诱导率为67.00%;生长素与细胞分裂素的添加浓度与搭配比例决定不定芽的分化速度,随着6-BA浓度的增加,其分化速度有加快的趋势;继代培养基采用MS+6-BA 2.0~3.0mg/L+NAA 0.05~0.10mg/L+蔗糖30g/L,琼脂7g/L,pH 5.8,增殖率为3.0~4.0;生根培养基采用MS+NAA 0.1mg/L+蔗糖30g/L,琼脂7g/L,pH 5.8,生根率达100%。
Different concentrations of 6-BA and NAA impact on the induction and proliferation and rooting of Philodendron'Nobel' were studied,by using hoot tip and node as the explants.The results indicated that explants derived from different part of the plant were found to induce shoot at different rate.The induction rates for the shoot tip and the node were 91.50% and 67.00% respectively.The differentiation rate of adventitious buds was determined by the concentration and proportion between 6-BA and NAA;The proliferation rate of adventitious buds increased along with the rise in the concentration of 6-BA.The culture medium MS+6-BA 2.0~3.0 mg/L+NAA 0.05~0.1 mg/L+sugar 30 g/L+agar 7 g/L at pH 5.8 was preferable for subculture proliferation,PC 3.0~4.0.The most favorable growth obtained was with MS+NAA 0.1 mg/L+sugar 30 g/L+agar 7 g/L,at pH 5.8,producing 100% rooting rate.
