应用DHPLC筛查黑白头发mtDNA控制区的异质性研究

摘要 目的探讨黑头发和白头发线粒体DNA(mtDNA)控制区的异质性。方法将人头发mtDNA控制区扩增成5个部分互相重叠的片段,利用已建立的DHPLC技术分析中国汉族正常人与Alzheimer病同个体黑头发和白头发mtDNA控制区的异质性。结果 50例中国汉族正常人同个体中黑头发和白头发mtDNA控制区的异质性为10%和20.4%,Alzheimer病黑头发和白头发mtDNA控制区的异质性为16.4%和30%。结论同个体中黑头发和白头发线粒体DNA(m tDNA)控制区异质性与衰老相关。 To explore relationship of Genetics of Heteroplasmy in the mtDNA Control Region in patients with Alzheimer diseases.Methods The genotyes of mtDNA mutation were determined by Denaturing High-Performance Liquid Chromatography(DHPLC) in 50 patients with Alzheimer disease and 50 healthy controls.Results There were siganificant differences in the frequenceies of mtDNA mutations between the patients and central groups(P〈0.05).Conclusion Genetics of Heteroplasmy in the mtDNA Control Region may be dependent risk factors for Alzheimer′s disease.
