摘要
采用ISSR分子标记技术对华仁杏Armeniaca cathayana D.L.Fu et al.3个杂交组合(A:H1×H2,B:H1×X1,C:H1×X2)的亲本及杂交一代(F1)进行了遗传分析。从100条ISSR引物中筛选出稳定多态的8条引物,共得到52个扩增位点,其中44个具有多态性,多态性位点百分率达84.62%。在华仁杏3对杂交组合中,组合B的F1子代同母本的遗传相似系数平均为0.6601,高于其亲本间的遗传相似系数0.6078,同父本的遗传相似系数平均为0.5882,低于其亲本间的遗传相似系数。组合A和组合C的F1子代同父本、母本之间的遗传相似系数平均值都低于其各自亲本间的遗传相似系数。同时3个杂交组合F1的有效等位基因数、期望杂合度、shannon多样性指数都高于两亲本间的。这表明华仁杏3个杂交组合的F1子代都产生了明显的的遗传变异,遗传多样性水平得到了提高。
Using ISSR molecular markers,the genetic variations among parents and F1 hybrids of three hybrid combinations(A:H1×H2,B:H1×X1,C:H1×X2)of Armeniaca cathayana D.L.Fu et al.were analyzed.Eight stable and polymorphic primers were sifted from 100 ISSR primers,which could get 52 amplification loci and 44 polymorphic loci,with polymorphism loci percentage 84.62%.In combination B,the average genetic similarity coefficient between F1 and maternal plant was 0.6601,which higher than that between their parent plants 0.6078,and the average genetic similarity coefficient between F1 and parternal plant was 0.5882,which less than that between their parent plants.In the other two hybrid combination A and C,the average genetic similarity coefficients between F1 and parent plants all were less than those between their parent plants respectively.Meanwhile the alleles,expected alleles,expected heterozygosities and shannon diversity indexes of F1 hybrids of three combinations all were higher than their parents respectively.This shows that the F1 hybrids of three combinations of A.cathayana had more genetic variations and more genetic diversity in DNA level.
