摘要
[目的]建立同时测定竹节参药材中3种主要化学成分(竹节参皂苷Ⅴ、竹节参皂苷Ⅳa和竹节参皂苷Ⅳ)的HPLC含量测定方法;在此基础上,考察湖北恩施竹节参各主要成分含量与栽培年限之间的动态变化。[方法]采用HPLC对不同生长年限的竹节参栽培品中竹节参皂苷Ⅴ、竹节参皂苷Ⅳa、竹节参皂苷Ⅳ的含量进行测定。色谱条件:色谱柱为Ultimate XB-C18色谱柱,(4.6 mm×250 mm,5μm);流动相为乙腈-浓度0.2%乙酸铵溶液(32∶68,V/V);流速为1.0 ml/min;检测波长203 nm,柱温30℃。[结果]竹节参皂苷Ⅴ、竹节参皂苷Ⅳa、竹节参皂苷Ⅳ的含量分别在0.06~4.00、0.04~3.00、0.04~3.00μg/ml浓度范围内线性关系良好(r1=0.999 6、r2=0.999 4、r3=0.999 5);平均加样回收率分别为100.61%、99.06%、101.12%,RSD分别为1.28%、1.63%、0.89%;竹节参中各主要成分含量随栽培年限变化较大。[结论]该方法简便、可靠,可用于测定竹节参各主要成分的含量;通过监测竹节参各主要成分的累积规律,可为制定竹节参规范化栽培措施提供依据。
[Objective] To establish HPLC method that could determine simultaneously the content of three main chemical constituents like P.japonicus saponins V,P.japonicus saponins IVa and P.japonicus saponins IV in P.japonicas medicinal materials,and to study the dynamic variation of three chemical constituents of Hubei Enshi P.japonicus in different cultivating years.[Method] The content of P.japonicus saponins V,P.japonicus saponins IVa and P.japonicus saponins IV in each sample of P.japonicas was determined by HPLC.The HPLC analysis was performed on a Diamonsil C18 column(250 mm×4.6 mm,5μm) with the detection wavelength at 203 nm.Following the injection of sample solution,a elution with acetonitrile-0.2% ammonium acetate(V/V,32∶68) as mobile phase was initiated at a flow rate of 1.0 ml/min.In addition,the column temperature was 30 ℃.[Result] The calibration curve of P.japonicus saponins V,P.japonicus saponins IVa and P.japonicus saponins IV were linear in the range of 0.06-4.00 μg(r1=0.999 6),0.04-3.00 μg(r2=0.999 4),0.04-3.00 μg(r3=0.999 5),respectively.The average recovery of the above 3 chemical constituents was 100.61%(RSD=1.28%),99.06%(RSD=1.63%),101.12%(RSD=0.89%),respectively.The content of the main constituents in P.japonicus differed greatly in different cultivating years.[Conclusion] The method is simple,reliable,and can be used for determining the content of the main constituents in P.japonicus.It could provide reference to formulate standardized cultivation measures for P.japonicus by monitoring the accumulation of its main constituents.
