摘要
[目的]制备可以识别双酚A(BPA)的多克隆抗体。[方法]通过双酚酸(DPA)与载体蛋白牛血清白蛋白(BSA)以及卵清白蛋白(OVA)偶联制备免疫抗原DPA-BSA和包被抗原DPA-OVA,以DPA-BSA免疫新西兰大白兔获得抗血清,并采用间接竞争酶联免疫吸附(ELISA)检测方法对抗体进行鉴定。[结果]经紫外光谱分析,DPA-BSA和DPA-OVA制备成功。抗血清效价最高达到1∶256 000,50%抑制率(IC50)达17.2 ng/ml。[结论]该研究为建立双酚A的快速筛选方法奠定了基础。
[Objective] The paper aimed to prepare polyclonal antibody that could recognize Bisphenol A.[Method] Diphenolic acid(DPA)was used to couple with carrier protein BSA and OVA for preparing immune antigen DPA-BSA and coating antigen DPA-OVA,and the New Zealand white rabbits were immunized with DPA-BSA to prepare antiserum.The antibody was determined by indirect competitive enzyme-linked immunosorbent assay(ELISA).[Result] UV-Spectroscopic analysis showed that DPA-BSA and DPA-OVA were successfully prepared.The antiserum titer was as high as 1∶ 256 000,and 50% inhibitory concentration was 17.2 ng/ml.[Conclusion] The study lays a foundation for developing rapid detection method of BPA.
