骨髓增生异常综合征和急性髓系白血病患者骨髓间充质干细胞的生物学特性

Biological Characteristics of Mesenchymal Stem Cells Derived From Bone Marrow of Patients with Myelodysplastic Syndrome or with Acute Myeloid Leukemia

摘要 目的研究骨髓增生异常综合征(MDS)和急性髓系白血病(AML)患者的骨髓间充质干细胞(MSC)的生物学特性。方法获取7例MDS和21例AML患者的骨髓标本,以6例正常成人的MSC为对照组。体外分离、培养和扩增MSC,显微镜观察MSC的形态和生长特性,免疫细胞化学染色和流式细胞计数进行表型鉴定,RT-PCR法检测MSC中IL-6、SDF-1、SCF及BCL-2的mRNA表达。结果 5/21例AML和1/7例MDS骨髓基质层生长不良,16/21例AML和6/7例MDS患者骨髓获得可传代的MSC具有相似的细胞形态和免疫表型,但AML和MDS的MSC在mRNA水平上表达IL-6和SDF-1细胞因子的量均显著高于对照组,且AML患者MSC的倍增时间明显长于MDS。结论 MDS和AML的MSC生物学特性与对照组比较存在异常。 Objective To study the biological characteristics of mesenchymal stem cells(MSC) derived from bone marrow of patients with myelodysplastic syndrome(MDS) or with acute myeloid leukemia(AML).Methods MSC were derived from bone marrow samples of 7 patients with MDS,21 patients with AML and 6 normal control subjects,respectively.MSC were cultured and expanded in vitro.The morphology and proliferation of MSC were observed under light microscope;the immuno-phenotype of MSC were determined by immunocytochemistry staining and flow cytochemistry.The expression levels of IL-6,SDF-1,SCF and BCL-2 were detected by half-quantitative reverse transcription and polymerase chain reaction(RT-PCR).Results MSC derived from 5 AML patients and 1 MDS patients grew poorly.The other MSC derived from the rest 16 AML patients and 6 MDS patients could grow and pass well,which presented with similar cell morphology and similar immuno-phenotype.The mRNA expression levels of IL-6 and SDF-1 in MSC established from AML patients and MDS patients were significantly higher than those of control group.The doubling time of the MSC derived from AML patients was significantly longer than that of the MSC derived from MDS patients and the control group.Conclusion There are abnormal biologic characteristics associated with the bone marrow-derived MSC of AML and MDS in comparison with normal control MSC.
