盐酸氨基乙酰丙酸介导的635/670nm联合波长下光动力学治疗的抗肿瘤效应 被引量:1

摘要 目的通过实时测定单态氧(singlet oxygen,1 O2)评估635nm和(或)670nm激光照射下盐酸氨基乙酰丙酸(5-aminolevulinic acid,5-ALA)介导的光动力学治疗(photodynamic therapy,PDT)抗肿瘤效果。方法以5-ALA为光敏剂,在单波长(635nm或670nm)及联合波长(635nm+670nm)下进行激光照射,利用新型高灵敏的检测系统测定PpIX溶液、肿瘤细胞(9L神经胶质肉瘤细胞和HeLa细胞)及HeLa负瘤裸鼠中的1 O2强度,H-E染色法检测肿瘤组织坏死深度。结果接受单一的635nm或670nm波长激光照射时,PpIX溶液1 O2强度在两者中无明显区别。635nm与670nm激光联合照射下,9L细胞的1 O2强度高于HeLa细胞;在两种细胞中670nm激光诱发1 O2强度均远远低于635nm激光。在HeLa细胞裸鼠皮下移植瘤中,两种波长激光联合照射产生一定的肿瘤坏死深度,但低于635nm单波长照射。结论在5-ALA介导的PDT中,670nm波长激光显示了一定的PDT疗效。
