摘要
目的:观察葡萄籽原花青素(grape seed proanthocyanidin extract,GSPE)对粘性放线菌生长、黏附和产酸的影响。方法:选用粘性放线菌ATCC15987为实验菌株。采用液体稀释法测量GSPE对粘性放线菌的最低抑菌浓度(minimum inhibitory concentration,MIC);菌株在含低于MIC 4个浓度的TPY液体培养基中厌氧培养,采用同位素闪烁计数法测细菌对唾液包被的羟基磷灰石(saliva coated hydroxyapatites,SHA)的黏附量(count per minute,CPM);调定液体培养基初始pH值为7.20,厌氧培养后测其终末pH值,并计算pH变化值(△pH)。结果:GSPE对粘性放线菌的MIC为8.00 mg/ml。1.00 mg/ml GSPE培养基中细菌CPM高于阳性对照组(P<0.05),与阴性对照组相比差异无显著性(P>0.05)。大于1.00 mg/ml GSPE的其余3个实验组均高于阳性对照组,但低于阴性对照组(P<0.05)。各实验组△pH均低于空白对照组(P<0.05)。CPM及△pH的值随GSPE浓度的增加而降低。结论:葡萄籽原花青素对粘性放线菌的生长、黏附和产酸有一定的抑制作用。
Objective: To observe the effect of grape seed proanthocyanidins extract(GSPE) on the growth,adhesion and acid production of Actinomyces viscosus(A.v).Methods: A.v ATCC 15987 was cultured in TPY medium containing various concentrations of GSPE.The minimum inhibitory concentration(MIC) was determined by the broth dilution method.The count per minute(CPM) of the bacteria to saliva coated hydroxyapatites(SHA) was detected by radioisotope scintillation counting.The initial pH of the medium was 7.2,and finally the rest pH was measured.Results: The MIC of GSPE against A.v was 8.00 mg/ml.The CPM in 1.00 mg/ml GSPE group was higher than that in the positive control group(P〈0.05),but there was no significant difference between experimental group and the negative control group(P〈0.05).The CPM values of other experimental groups were higher than that in the positive control group and lower than in the negative control group(P〈0.05).The △pH of experiment groups were lower than that of the control group(P〉0.05).The CPM and the △pH value decreased with the increasing of GSPE concentration.Conclusion: GSPE can inhibit the growth,acid production and adhesion of A.v.
