摘要
目的探讨眼针治疗急性脑缺血再灌注损伤的机制。方法应用线栓法复制急性脑缺血再灌注损伤大鼠模型,采用眼针治疗,于再灌注后3 h进行神经功能缺损评分,采用免疫组化、Western印迹、实时荧光定量聚合酶链反应(RQ-PCR)方法测定脑组织脑源性神经因子(BDNF)蛋白及mRNA表达的变化。结果与模型组比较,眼针组大鼠神经功能缺损评分明显下降,脑组织BDNF蛋白及mRNA表达明显上调(P<0.05)。结论眼针具有明显改善大鼠脑缺血再灌注损伤的作用;其机制与上调脑组织BDNF表达有关。
Objective To investigate the effect of eye acupuncture therapy on cerebral brain-derived neurotrophic factor(BDNF) expres sion in rats with cerebral ischemia-reperfusion injury and the related mechanis m. Methods Rat model of cerebral ischemia-reperfusion was e stablished by suture method.The methods of immunohistochemistry staining,West ern-blotting and RQ-PCR were taken to detect the alteration of the BDNF protei n and BDNF mRNA in rat brains after eye acupuncture therapy. Results Compared with the model group,the neurologic imp airment sco re of the eye-acupuncture group was decreased obviously and the expression of t h e BDNF protein and BDNF mRNA in rat brains after eye acupuncture therapy was str ongly increased(P0.05). Conclusions Eye acupuncture therapy can reduce the ce rebral ischemia-reperfusion injury evidently by increasing the expression of th e cerebral BDNF protein.
