人脐血间质干细胞向神经元样细胞诱导分化促进大鼠脊髓损伤神经恢复 被引量:2

Induction differentiation of transplantated human cord blood MSCs into neuro-like cells improves recovery of neurological function after spinal cord injury in rats

摘要 目的观察移植人脐血间充质干细胞诱导分化的神经元样细胞是否可以促进大鼠脊髓损伤的功能恢复。方法 30只SD大鼠随机分成移植组和对照组。建立NYU脊髓损伤模型。移植组移植Hoechst标记的源于脐血间充质干细胞在体外培养、扩增,向神经元样细胞分化的细胞,并用免疫荧光标记方法检测神经元特异性标记物。对照组注射PBS液。采用BBB评分标准对两组所有大鼠术前和术后24 h,1、2、3、4、5周进行运动功能评价,组织学和免疫组化分析移植细胞的定位和分化情况。结果经诱导后的脐血间充质干细胞表达神经丝蛋白和神经元特异性烯醇化酶,移植后在脊髓病理切片中出现Hoechst标记的移植细胞,5周后移植组功能恢复比对照组明显(P<0.05)。结论移植人脐血间充质干细胞诱导分化为神经元样细胞可以促进鼠脊髓损伤的功能恢复。 Objective To study whether induction differentiation of transplantated human umbilical cord blood(HUCB) mesenchymal stem cells(MSCs) into neuro-like cells can provote neurological functional recovery after spinal cord injury in rats. Methods After establishment of NYU spinal cord injury model,30 SD rats were randomly divided into two groups.The rats in group A were transplanted MSCs of HUCB,which underwent induction differentiation into regenerate neuron like cells and labed by Hoechst in vitro.The rats in group B were injected with PBS as the controls.The neurological function was determined using the modified BBB score before and at 24h and 1,2,3,4 and 5 weeks after spinal cord injury.The distribution and differentiation of transplanted cells in rat spinal cord were evaluated by histological and immnuhistochemical analysis. Results The induced MSCs expressed neurofilament(NF) and neuron-specific enolase(NSE).Moreover,Hoechst-reactive transplantable cells were found to be survival in rat spinal cord.Functional recovery determined by modified BBB score was significantly improved in group A than that in group B(P0.05). ConclusionInduction differentiation of transplantated MSCs of HUCB into neuro-like cells can provote neurological functional recovery after spinal cord injury in rats.
