放疗耐受星形胶质细胞瘤的蛋白组学分析

A proteomic analysis of radioresistant astrocytomas

摘要 目的分析不同放疗敏感性星形胶质细胞瘤蛋白表达特点。方法以放疗后预后为依据,将WHOⅡ级星形胶质细胞瘤分为放疗敏感组和耐受组。应用二维液相色谱-质谱联用法分析胶质瘤蛋白表达,鉴定两组间差异表达蛋白。对差异蛋白磷酸甘油酸激酶1进行Western blot验证。结果磷酸甘油酸激酶1在胶质瘤放疗耐受组中过表达。结论磷酸甘油酸激酶1增多可能与胶质瘤放疗耐受的发生相关。 Objective To analyze the features of protein expression of radioresistant astrocytomas. Methods On the basis of proteome patterns of the radiosensitivity(WHO Ⅱ) the astrocytomas were divided into two groups of radiosensitivity and radioresistance.The protein expression was analyzed by two-dimensional liquid chromatography tandem mass spectrometry(2D-LC-MS/MS).Western blot was used to quantify phosphoglycerate kinase 1(PGK1). ResultsPGK1 was overexpressed in the radioresistant astrocytomas. Conclusion PGK1 might be correlated with glioma radioresistance.
