柯萨奇病毒心肌病动物模型研究

摘要 目的研究反复增量柯萨奇病毒B3m(CVB3m)感染对小鼠心肌的影响,建立病毒性心肌病动物模型。方法 80只3~4周龄雄性Balb/c小鼠,随机分为心肌病组、单次感染组和正常对照组,于首次感染病毒后第120 d处死小鼠,应用阻抗微分法测定心输出量,病理及组织化学技术分析心肌病理损伤和胶原系统改变,计算胶原容积分数。结果心肌病组小鼠死亡率增高,心脏重量增加,心功能下降,心肌胶原容积分数高于单次感染组和对照组(P<0.05),病理学特征为基质胶原明显增生重建。结论反复增量病毒感染产生了明显的心室重构,符合心肌病特点,可做为病毒性心肌病动物模型进一步研究。 Objective To study the influence of myocardium by repetitive increased CVB3m infection and establish virus cardiomyopathy model.Methods 80 male 3~4 week Balb/c mouse were divided cardiomyopathy group,single infected group and control group randomly.The mice were sacrificed on 120 days post first infection.Impedance differentiation was used to calculate cardiac output(CO).The changes of myocardial pathology and collagen were used pathology,histochemistry technique.And the collagen volume fraction was counted.Results In cardiomyopathy group,the death rate and the heart weight were increased,the cardiac function were decreased.And the collagen volume fraction was higher than the other groups(P0.05).The pathologic feature is the significant proliferation and reconstruction of the extracellular matrix collagen.Conclusion Repetitive CVB3m infection make obviously ventricular remodeling.The pathology is consistent with cardiomyopathy.The model could be used as cardiomyopathy model to future investigate.
