摘要
目的探讨重组血管紧张素转换酶2(ACE2)基因对血管平滑肌细胞前纤维蛋白-1(Profilin-1)表达及细胞增殖的影响。方法培养人脐动脉平滑肌细胞(HUASMC),用重组ACE2基因(rACE2)干预,进行血管紧张素Ⅱ(AngⅡ)刺激实验,分别用MTT法、实时定量PCR及Western blot检测HUASMC增殖与Profilin-1表达。结果与对照组相比,AngⅡ(100 nmol/L)刺激6 h后,HUASMC中Profilin-1表达明显增高(3.50±0.30 vs 1.00±0.10,P<0.05)。重组ACE2基因干预显著抑制上述增加(1.73±0.12 vs 3.50±0.30,P<0.05)。结论 ACE2基因过表达可明显逆转HUASMC中AngⅡ诱导的Profilin-1表达上调。
Objective To investigate the effects of the recombinant angiotensin-converting enzyme 2 (rACE2) gene on Profilin-1 expression and cell proliferation in vascular smooth muscle cells. Methods The cultured human um- bilical arterial smooth muscle cells (HUASMC) were stimulated by Ang II in the presence or absence of rACE2 gene, HUASMC proliferation and Profilin-1 expression level were determined with MTF, real-time PCR and West- HUASMCs (3.50 ±0. 30 vs 1. 00 ±0. hours, which was inhibited by rACE2 10 ; n = 5 - 6, P 〈 0. 05 ) by stimulating with Ang 1I ( gene intervention (1.73 ±0. 12 vs 3.50 ±0. 30; n =5,6, 100 nmol/L)for 6 P 〈 0. 05). Condusions ACE2 gene over-expression contributes to the reversion of the Ang II-induced Profilin-1 expression and proliferation in human umbilical arterial smooth muscle cells.
