摘要
采用ISSR分子标记技术,对广东省内分布的能源植物五节芒(Miscanthus floridulus)13个野生居群共215个个体的遗传多样性进行了研究。9个ISSR引物共扩增到了81个位点,其中76个是多态性位点。结果表明,广东五节芒的遗传多样性水平很高,物种水平上,多态位点百分率(PPB)为93.83%,Nei's基因多样性指数(He)为0.3969,Shannon's信息指数(Hsp)为0.5724;居群水平上,PPB=81.91%,He=0.3015,Hpop=0.4460。居群间的遗传分化低于居群内的遗传分化,居群间遗传多样性分化系数(Gst)为0.2379,居群间基因流(Nm)为1.6018。AMOVA分析表明,广东五节芒居群间的遗传变异占总变异的20.44%,居群内的遗传变异占79.56%。居群的地理分布和遗传多样性分布没有直接的相关性。Mantel检测表明,居群遗传距离和地理距离之间无显著的相关关系(r=-0.2811,P=0.9590),居群遗传多样性无明显的地域性分布格局。
The genetic diversity of 215 individuals of Miscanthus floridulus from 13 natural populations distributed in Guangdong Province was investigated by using intersirnple sequence repeat (ISSR) techniques. A total of 81 unarvbiguous and repetitious bands were scored by 9 primers. The resuks showed that the genetic diversity of Mr. flotidulus was high, at species level, percentage ofpolyrrorphic loci (PPB) was 93.83%, Nei's gene diversity (He) for 0.3969, and Shannon's inforrmtion index (Hsp) for 0.5724; while at population level, PPB = 81.91%, He = 0.3015 and Shannon's inforrmtion index (Hpop) = 0.4460. The coefficient of genetic differentiation among populations (Cat) was 0.2379, and gene flow among populations (Nm) was 1.6018. The AMOVA analysis showed that 79.56% of the total genetic variation within populations and the rest (20.44%) partitioned among populations. No significant correlation was found between geographical and genetic distance (r=-02811, P=0.9590).
