摘要
目的:探讨不同治疗周期脉冲染料激光照射对增生性瘢痕动物模型伤口愈合过程中成纤维细胞增殖与凋亡的影响。方法:大耳白兔10只,建立兔耳腹侧面增生性瘢痕模型,对比研究在瘢痕形成过程中不同治疗周期(1周治疗组与2周治疗组)脉冲染料激光照射对兔耳瘢痕成纤维细胞的影响,采用免疫组化方法检测增殖细胞核抗原(PCNA)蛋白表达和细胞凋亡的原位检测。结果:兔耳增生性瘢痕经不同治疗周期(1周治疗组与2周治疗组)脉冲染料激光照射后,按不同时间段取材进行免疫组化染色比较,两组无明显差异。结论:不同治疗周期脉冲染料激光照射均可抑制兔耳增生性瘢痕成纤维细胞的增殖过程,诱导细胞凋亡。但从减轻医生工作强度及减轻病患治疗费用角度考虑,利用脉冲染料激光治疗增生性瘢痕2周一次较为合适。
Objective To investigation the effect of pulse dye laser with different treatment cycle on proliferative activity and apoptosis of fibroblasts in the hypertrophic scarring models in rabbit ears.Methods 10 white rabbits were used to establish the models of hypertrophic scar in ears.To study the change of fibroblasts pulse dye laser irradiation with different treatment cycle(one-week treatment control and two-week treatment control) and compare with the two controls during the process of forming hypertrophic scars.The expression of proliferation cell antigen(PCNA) protein was measured by immunohistochemistry technique,and the apoptosis was measured by TUNEL technique.Results The samples were taken in different treatment cycle after pulse dye laser irradiation treatment.The expression of PCNA and apoptosis don't have obvious distinction.Conclusion Pulse dye laser irradiation can inhibit the proliferation of fibroblasts and acceleration apoptosis in hypertrophic scars of rabbit ears and has significant effect on hypertrophic scars,but take the workload of the doctor and expense of the patient into consideration,it is proper to apply pulse dye laser treatment once in a fortnight.
