膀胱尿路上皮癌中DNMT1的表达及其临床意义

摘要 目的研究DNA甲基转移酶1(DNMT1)在膀胱尿路上皮癌(BUC)中的表达,探讨DNMT1与膀胱癌生物学行为之间的关系。方法采用RT-PCR法和Western-blot技术,检测60例膀胱尿路上皮癌及12例正常膀胱黏膜组织中DNMT1的表达;并结合临床资料进行分析。结果①膀胱癌组织中DNMT1蛋白和mRNA的表达水平分别为0.628±0.245和0.774±0.083,显著高于正常膀胱黏膜组织的0.126±0.028和0.175±0.101(P=0.000);②DNMT1在膀胱癌中的表达水平与肿瘤的临床分期、分化程度、有无淋巴结转移等生物学行为显著相关。结论 DNMT1在膀胱尿路上皮癌中过度表达,在膀胱癌的发生发展中起重要作用。 Objective To investigate the expression and clinical significance of the DNA methltransferasesl (DNMT1) in the bladder urothelial carcinoma (BUC). Methods Expressions of DNMT1 in 60 specimens of bladder urothelial carcinoma and 12 specimens of normal bladder tissues were detected with RT-PCR and Western-blot. The results were analyzed com- bined with clinical data. Results The expressions of DNMT1 protein and mRNA in bladder cancer were 0. 628 ±0. 245 and 0. 774 ± 0. 083, which were significantly higher than those of normal bladder epithelium, 0. 126 ± 0. 028 and 0. 175 ± 0. 101 (P =0. 000). The expressions of DNMT1 in neoplastic tissues were positively related to the clinical stage, tumor grade and the lymph node metastasis of cancer cells. Conclusions DNMT1 overexpression in bladder cancer may be associated with tumor progression.
